Turning RacGAP into a RhoGAP
نویسنده
چکیده
e already knew the players—MgcRacGAP, Aurora B, and RhoA—and that knocking out any one of them caused failure of cytokinesis; but it wasn’t clear how they were connected. Now, it appears that Aurora B phosphorylates the GAP domain of MgcRacGAP, allowing it to turn its GAP activity toward RhoA, according to data from Yukinori Minoshima, Toshiyuki Kawashima, Toshio Kitamura (University of Tokyo, Tokyo, Japan), and colleagues. When they first isolated MgcRacGAP, the group found that most of its activity was directed toward Rac1 and Cdc42, but there were hints—although inconsistent ones—that it also had activity toward Rho proteins. In the new work, Kitamura and colleagues found that, late in mitosis, Aurora B, RhoA, and MgcRacGAP congregate at the midbody, where Aurora B phosphorylates a serine in the GAP domain of MgcRacGAP. The phosphorylated GAP protein then stimulates the GTPase activity of RhoA, converting GTP-bound active RhoA to GDP-bound inactive RhoA, and promotes the completion of cytokinesis and cell division. Overexpression of a phosphorylation-deficient mutant of MgcRacGAP blocks RhoA activity and results in polyploid cells. “This is the first demonstration that a modification of a GAP changes its target specificity,” says Kitamura. “Biologically the interesting thing is that in the beginning of cell division MgcRacGAP probably works through Rac1 and Cdc42, which function in mitotic spindle formation. Then during the late stage of cell division and cytokinesis, MgcRacGAP converts itself to a RhoGAP and exerts itself through RhoA.”

p190RhoGAP has cellular RacGAP activity regulated by a polybasic region.
p190RhoGAP is a GTPase-activating protein (GAP) known to regulate actin cytoskeleton dynamics by decreasing RhoGTP levels through activation of the intrinsic GTPase activity of Rho. Although the GAP domain of p190RhoGAP stimulates the intrinsic' GTPase activity of several Rho family members (Rho, Rac, Cdc42) under in vitro conditions, p190RhoGAP is generally regarded as a GAP for RhoA in the ce...
متن کاملA screen for modifiers of RacGAP(84C) gain-of-function in the Drosophila eye revealed the LIM kinase Cdi/TESK1 as a downstream effector of Rac1 during spermatogenesis.
In Drosophila, RotundRacGAP/RacGAP(84C) is critical to retinal organisation and spermatogenesis. We show that eye-directed expression of RacGAP(84C) or its GTPase activating protein (GAP) domain induces a dominant rough eye phenotype which we used as a starting point in a gain-of-function screen to identify new partners of RacGAP(84C). Proteins known to function in Ras, Rho and Rac signalling w...
متن کاملTum/RacGAP functions as a switch activating the Pav/kinesin-6 motor.
Centralspindlin is essential for central spindle and cleavage furrow formation. Drosophila centralspindlin consists of a kinesin-6 motor (Pav/kinesin-6) and a GTPase-activating protein (Tum/RacGAP). Centralspindlin localization to the central spindle is mediated by Pav/kinesin-6. While Tum/RacGAP has well-documented scaffolding functions, whether it influences Pav/kinesin-6 function is less wel...
متن کاملQuantitative proteome analysis of pluripotent cells by iTRAQ mass tagging reveals post-transcriptional regulation of proteins required for ES cell self-renewal.
Embryonic stem cells and embryonal carcinoma cells share two key characteristics: pluripotency (the ability to differentiate into endoderm, ectoderm, and mesoderm) and self-renewal (the ability to grow without change in an untransformed, euploid state). Much has been done to identify and characterize transcription factors that are necessary or sufficient to maintain these characteristics. Oct-4...
متن کاملCoordination of Rho and Rac GTPase Function via p190B RhoGAP
The Rac GTPase regulates Rho signaling in a broad range of physiological settings and in oncogenic transformation [1-3]. Here, we report a novel mechanism by which crosstalk between Rac and Rho GTPases is achieved. Activated Rac1 binds directly to p190B Rho GTPase-activating protein (RhoGAP), a major modulator of Rho signaling. p190B colocalizes with constitutively active Rac1 in membrane ruffl...
